Peptide Synthesis Overview¶
TL;DR¶
Peptide synthesis is the production of custom peptide chains through either solid-phase (SPPS) or liquid-phase (LPPS) methods. SPPS is the dominant technology, offering automation, high yield, and scalability from milligrams to kilograms.
Peptide synthesis is the chemical process of assembling amino acids into defined sequences to create peptide chains. The most widely used method is Solid-Phase Peptide Synthesis (SPPS), first developed by Bruce Merrifield in 1963.

- Reaction Time: 30 min to several hours per coupling
- Excess Reagents: 2–5 fold molar excess of amino acids
- Solvent: DMF or NMP as standard reaction medium
- Mixing: Nitrogen bubbling or mechanical agitation
