Lyophilization¶
Freeze-Drying of Peptides¶
Purpose¶
Convert liquid peptide solution into stable dry powder by freezing and sublimation under vacuum.
Lyophilization Process¶
graph TD
A[Peptide Solution] --> B[Freezing];
B --> C[Primary Drying
Sublimation];
C --> D[Secondary Drying
Desorption];
D --> E[Final Product]; Step 1: Freezing¶
Objective¶
Convert liquid water to ice, concentrating the peptide in a glassy or crystalline matrix.
Parameters¶
| Parameter | Typical Value |
|---|---|
| Freeze temperature | -40 to -80 °C |
| Freeze rate | 0.5–2 °C/min |
| Annealing | Optional, -20 °C for 1–2 h |
Considerations¶
- Fast freezing: Smaller ice crystals, larger surface area
- Slow freezing: Larger crystals, faster primary drying
- Annealing: Promotes ice crystal growth, reduces drying time
Step 2: Primary Drying¶
Objective¶
Sublime ice directly to vapor under vacuum.
Parameters¶
| Parameter | Typical Value |
|---|---|
| Chamber pressure | 0.1–0.5 mbar |
| Shelf temperature | -20 to -10 °C |
| Product temperature | Maintain below collapse temp |
| Duration | 12–48 hours |
Collapse Temperature¶
- Typically -20 to -40 °C for peptides
- Varies with formulation (bulking agents raise collapse temp)
- Critical quality parameter — product must not melt
Endpoint Determination¶
- Product temperature approaches shelf temperature
- Pressure rise test (Pirani vs. capacitance manometer)
Step 3: Secondary Drying¶
Objective¶
Remove bound water from the peptide cake.
Parameters¶
| Parameter | Typical Value |
|---|---|
| Shelf temperature | 20–40 °C |
| Pressure | ≤0.05 mbar |
| Duration | 4–12 hours |
Target Residual Moisture¶
- Peptides: <5% (manufacturing spec)
- Desirable: <2% for long-term stability
Lyophilizer Components¶
| Component | Function |
|---|---|
| Vacuum pump | Maintain low pressure |
| Refrigeration | Cool shelves and condenser |
| Condenser trap | Trap water vapor (-50 to -80 °C) |
| Shelf system | Heat/cool product temperature |
| Control system | Recipe programming and monitoring |
Quality Considerations¶
Cake Appearance¶
- Acceptable: Elegant, intact cake, white to off-white
- Defects: Cracking, shrinkage, melt-back, collapse
Common Issues¶
| Issue | Cause | Solution |
|---|---|---|
| Cake collapse | Temperature above collapse point | Slow ramp, lower temp |
| High residual moisture | Insufficient secondary drying | Extend secondary drying |
| Partial melting | Vacuum failure or pressure rise | Check seals, maintain vacuum |
Storage After Lyophilization¶
- Container: Air-tight vials or bags with desiccant
- Temperature: -20 °C (long-term), 4 °C (short-term)
- Protection: Light-sensitive peptides require amber vials
-
Shelf life: 2–5 years at -20 °C under dry conditions
